血小板衍生生长因子-BB体外促进破骨细胞样细胞形成
Plateket-derived growth factor (DDGF)-BB stimulates formation of osteoclast-like cells in vitro
摘要
目的 获得大量和高纯度破骨细胞样细胞。方法 从急性单核细胞性白血病 (M5a亚型 )患者骨髓中分离前体单核细胞 ,与血小板衍生生长因子 (PDGF) -BB共同培养 ,获得多核细胞 ;利用免疫电镜、免疫荧光、扫描电镜等技术鉴定多核细胞的生物学特性。结果 多核细胞表达抗酒石酸酸性磷酸酶 (TRAP) ,细胞膜具有Vitronectin受体 ,能够形成骨吸收陷窝。结论 所获得的多核细胞是破骨细胞样细胞 ,PDGF -BB能够促进前体破骨细胞向破骨细胞转化。
