摘要
在pH 3.6的Clark Lub′s缓冲溶液中 ,四磺基锰酞菁 (MnTSPc)和蛋白质的共振瑞利散射 (RRS)均十分微弱。当两者形成复合物时能使RRS信号急剧增强 ,在 330~ 370nm范围内呈现较高的散射强度 ,其最大散射波长为 344nm。各种蛋白质 ,包括牛血清白蛋白 (BSA)、人血清白蛋白 (HSA)、γ 球蛋白 (γ IgG)及卵蛋白(OVB)的线性范围分别为 0 .1~ 3.0、0 .0~ 1.6、0 .0 5~ 2 .0及 0 .0~ 1.0mg/L ;相应的检出限分别为 5 .7、8.3、13和 13μg/L。用此法对人血清样品进行了测定 。
At pH 3.6, the interaction of manganese-tetrasulfonatophthalocyanine with proteins results in enhanced Rayleigh light scattering signals at 344 nm. Proteins including bovine serum albumin, human serum albumin, human gamma-immunoglobulin, and ovalbumin, can be determined under optimum conditions', the,linear ranges of the calibration curves were 0.0similar to1.6 mg/L for human serum albumin, 0.05similar to2.0 mg/L for human gamma-IgG, 0.1similar to3.0 mg/L for the bovine serum albumin and 0.0similar to1.0 mg/L for ovalbumin, the corresponding detection limits were 5. 7, 8.3, 13 and 13 mug/L, respectively. The method has been applied-to the analysis of total protein in human serum samples collected from the hospital and the results are satisfactory.
