摘要
目的 :建立祛风活络丸中士的宁和马钱子碱的反相高效液相分析法 ,为该药质量控制提供依据。方法 :采用Tech sphese C18ODS色谱柱 (4.6mm× 2 5 0mm ,5 μm) ;乙腈 甲醇 0 .0 5mol·L-1KH2 PO4溶液 三乙胺 (2 5∶19∶5 5∶1,磷酸调pH至3 .5 )为流动相 :流速 1mL·min-1;检测波长 2 5 4nm。结果 :士的宁在 0 .3~ 3.6 μg(r =0 .9999)、马钱子碱在 0 .2~ 2 .4 μg(r =0 .9996 )范围内呈良好的线性关系 :士的宁回收率为 97.6 % ,RSD =2 .0 % ;马钱子碱回收率为 10 1.0 2 % ,RSD =1.4 %。结论 :本方法准确可靠 ,简便灵敏 ,重现性好 ,可用于祛风活络丸中士的宁和马钱子碱的质量控制。
Objective: To establish a method for the determination of strychnine and brucine in Qufenghuoluo pills by RP-HPLC to provide a foundation for quality control of this agent. Method: Techsphese-C 18 ODS colum ( 4.6 mm×250 mm, 5 μm) was used, mobile phase consisted of acetonitril-methanol- 0.05 mol·L -1 KH 2PO 2 solution-triethylamine, pH was adjusted to 3.5, the flow rate was 1 ml·min -1,wavelength for assay was at 254 nm. Result: Strychnine and brucine presented a good linear correlation within a range of 0.3- 0.36 μg(r= 0.999 9) and 0.2- 0.24 μg(r= 0.999 6),respectively. The recovery of both components were 97.6% (RSD= 2.0%) and 101.02%(RSD= 1.4%), respectively. Conclusion: This method is definite, reliable, simple, and a good repeatibility, and can be used for quality control of strychnine and brucine in Qufenghuoluo pills.
