人横纹肌肉瘤中微管蛋白的表达

Study on Microtubulin of Human Rhabdomyosarcoma

目的 研究人横纹肌肉瘤中细胞骨架成分微管的改变。方法 采用免疫组化 SP法 ,对 70例人横纹肌肉瘤石蜡组织切片中微管蛋白的表达进行研究 ;应用激光共聚焦显微镜对三种不同转移潜能的横纹肌肉瘤细胞系 (PL A- 80 2 ,A6 73,RD)中微管进行免疫荧光染色研究。结果 人横纹肌肉瘤石蜡组织切片中普遍存在微管蛋白的低表达。肿瘤组织学分级 级 microtubulin- β的低表达细胞为 5 2 .38% , 级为 85 .0 0 % (P<0 .0 5 )。有转移、复发的 32例中 ,microtubulin- β的低表达细胞为 87.5 0 % ;无转移、复发的 38例中 ,其低表达细胞为 5 5 .2 6 % (P<0 .0 5 )。在三种人横纹肌肉瘤细胞系中 ,PL A- 80 2细胞的微管发育最不完善 ;RD细胞中微管形态接近正常肌母细胞 ;A6 73细胞微管发育程度居中 (P<0 .0 5 )。结论 人横纹肌肉瘤中 ,普遍存在微管蛋白的低表达。微管蛋白的减少和微管的发育不完善与肿瘤细胞的转移潜能 ,肿瘤的组织学分级和转移复发有关。

Objective To detect the expression of microtubules in human rhabdomyosarcoma. Methods Expression of microtubulin-β in 70 tissue slides from human rhabdomyosarcoma was detected by immunohistochemistry. Immunofluorescent staining was used to examine microtubules in PLA-802, A673 and RD under laser scanning confocal microscope. Results Expression of microtubulin-β in human rhabdomyosarcoma tissue was widely decreased. The low expression rate of microtubulin-β in histological grade-Ⅰtumor was 52.38% , and that in histological grade-Ⅲ tumor 85% (P<0.05 ). The low expression rate of microtubulin-β in 32 cases with metastasis and recurrence was 87.5%, and that in 38 cases without metastasis and recurrence 55.26% . Microtubules in the three cell lines showed different degrees of maldevelopment. The most severe degree was observed in PLA-802, and the less severe degree in RD (P<0.05). Conclusion The microtubulin shows extensive low expression in human rhabdomyosarcoma. The decrease and hypogenesis of microtubules in human rhabdomyosarcoma have relationship with tumor's histological grade, malignant degree, metastasis, and recurrence.
