摘要
目的 研究苯丙酸诺龙对烧伤大鼠肝细胞白蛋白与创面成纤维细胞前胶原蛋白 m RNA表达的影响。方法 采用随机对照设计 ,使用苯丙酸诺龙治疗 2 0 %体表面积深 度烫伤大鼠 ,分别于伤后第 4、7、14、2 1d处死 ,留取肝组织与创面肉芽组织 ,RNA一步法分别提取二者总 RNA,RT- PCR一步法扩增检测白蛋白与 α1( )前胶原蛋白 m RNA表达水平。结果 实验组肝脏白蛋白 m RNA与肉芽组织 αl( )前胶原蛋白 m RNA表达水平明显提高 ,在各时间点均明显高于同期对照组 (P<0 .0 5 ) ,伤后第 7、14 d为 m RNA快速增长时期 (P<0 .0 1)。结论 苯丙酸诺龙能有效上调肝脏白蛋白 m RNA与肉芽 α1( )前胶原蛋白 m RNA的表达水平 ,有利于蛋白合成与逆转负氮平衡 ,从整体与局部促进创面愈合。
Objective To assess the mechanism for the effect of nandrolone phenypropionate (NP) on hepatic albumin mRNA and granulational αl(Ⅰ)procollagen mRNA in burned rats with the aim to underpin the clinical application of anabolic steroids. Methods Thirty-two Wistar rats with a deep second-degree cutaneous burn of 20% total body surface area were randomly divided into two groups to receive either 5 mg/kg NP(NP group) or normal saline as placebo(control group) every other day. The expression copy quantities of albumin-mRNA in liver tissue and α1(Ⅰ)procollagen mRNA in granulation wound were measured by quantitative fluorescent RT-PCR respectively on the post-burned days 4, 7, 14 and 21. Results The expression levels of albumin-mRNA and α1(Ⅰ) procollagen mRNA in NP group were much higher than those in control group. The 7th and 14th days were the periods in which the albumin-mRNA and α1(Ⅰ)procollagen mRNA expression had been increasing obviously (P<0.01). Conclusion Nandrolone phenylpropionate could effectively up-regulate the expression of albumin-mRNA in liver tissue and the α1(Ⅰ)procollagen mRNA in granulation wound.
