气道嗜酸性粒细胞摄取吸入性颗粒抗原的研究 被引量:1

摘要 目的 :通过小鼠哮喘模型观察气道嗜酸性粒细胞 (EOS)能否摄取吸入的颗粒性抗原。方法 :予 6周龄 BAL B/ c雌性小鼠腹腔注射鸡卵清蛋白 (OVA)使其致敏 ,行 OVA (若丹明标记 )雾化吸入刺激后进行支气管肺泡灌洗 ,收集支气管肺泡灌洗液进行 EOS分离提纯 ,将纯化后的 EOS涂片后在荧光显微镜下观察其是否摄取红色标记的 OVA。结果 :荧光镜下清楚地观察到气道 EOS摄取了吸入的红色标记的 OVA,摄取了 OVA的 EOS形态完整。结论 :EOS在气道表面遭遇外来抗原后 ,可摄取经气道吸入的颗粒性抗原 ,这一重要功能使 EOS完成了作为抗原提呈细胞 (APC)所必需的首要任务。 Objective:To investigate whether or not eosinophils could phagocytize particular antigens in airways.Methods:After being sensitized intraperitonaelly and inhaled with OVA, 6-week-old BALB/c mice were lavaged to obtain eosinophils. Then, the eosinophils were purified and smeared on the slice and observed by microscopy to identify the ingestion of antigens.Result:Eosinophils could phagocytize the labeled OVA.Conclusion:Eosinophils within the lumina of airways could phagocytize particular antigens inhaled, These probably provid a new way to treat asthma.
