变形链球菌表面蛋白和葡糖基转移酶基因疫苗免疫防龋动物实验:体质量影响研究 被引量:3

摘要 目的 :了解变形链球菌基因疫苗单独及联合免疫对定菌鼠体质量的影响。方法 :2 8d龄Wistar大鼠 3 6只 ,随机分为pcDNA3 pac组、pcDNA3 gtfB组、pcDNA3 pac联合pcDNA3 gtfB组、变形链球菌灭活全菌阳性对照组、pcDNA3空载体阴性对照组和PBS液空白对照组 ,进行 3次双侧颌下腺腺周注射免疫 ,建立定菌鼠模型及诱龋实验 3个月 ,于实验开始和结束时测定体质量。结果 :基因疫苗免疫组体质量与 pcD NA3及PBS组无显著差异 (P >0 .0 5) ,而灭活全菌细胞免疫组体质量显著下降 (P <0 .0 1)。结论 :pcD NA3 gtfB和pcDNA3 pac对大鼠体质量无不良影响 。 Objective: To observe the influence of gene vaccines p cDNA3-pac and pcDNA3-gtfB of Streptococcus mutans (S.mutans) on weight of gnotobiotic rats by submandibular gland-target injection immunization. Methods: 36 Wistar rats of 28-day-old were equally divided into 6 groups.The rats in the 3 experimental groups were vaccinated with pcDNA3-p ac (group A), pcDNA3-gtfB(group B) or pcDNA3-pac combined with pcDNA3-gtfB(gr oup C) respectively,The rats in one positive control were vaccinated with ina ctive whole cell of S.mutans JBP(group D),those in negative and blank contr ols were injected with the vector pcDNA3 (group E) and 0.1 mol/L PBS buffer (gr oup F) respectively. Each rat was administrated with 100 μg vaccines or control materials by submandibular gland-target injection and vaccinated three times at 1~2 week intervals. All rats were fed with cariogenic diet 2000# for 3 month s.Body weight of the rats was measured before and after the 3-month experiment. Results: At the beginning of the experiment the body weight (g) of the rats in group A,B, C,D,E and F was 7.3±0.27,7.0±0.46, 7.2±0.76, 7.3±0.33,7.5±1.72 and 7.2±0.6 8 respectively (P>0.05);by the end of the experiment that was 24.5±1.05,28. 3±1.29,26.6±1.19,10.2±1.81, 26.3±1.54 and 27.3±1.38 respectively (D vs each of the other groups P<0.01,among the other groups P>0.05). Conclusion: Gene vaccines pcDNA3-pac and pcDNA3-gtfB have no unfavo rable influence on weight of gnotobiotic rats,while the inactive whole cell vac cine has.
