益气活血通腑法对类2型糖尿病大鼠骨骼肌GLUT4表达的影响 被引量:4

摘要 目的 :研究益气活血通腑中药 (加味桃核承气汤 )对类 2型糖尿病大鼠的作用及骨骼肌葡萄糖转运蛋白 4 (GLUT4 )mRNA表达的影响。方法 :采用低剂量链脲佐菌素 (STZ)腹腔注射加高热量饲料喂养导致大鼠类 2型糖尿病模型 ,灌胃给药 4周 ,检测糖耐量、空腹血清胰岛素及骨骼肌GLUT4mRNA表达量的变化。结果 :造模后大鼠注射葡萄糖后 30、6 0、12 0min血糖显著升高 ,血清空腹胰岛素水平无明显变化 ,骨骼肌GLUT4mRNA表达量显著降低。给药 4周后 ,各药对空腹血清胰岛素水平和注糖后 30、6 0min血糖无明显影响 ,但中药低剂量、二甲双胍、罗格列酮可使注糖后12 0min血糖下降 ;罗格列酮尚可降低空腹血糖。二甲双胍可使骨骼肌GLUT4mRNA表达量明显升高 ,中药高剂量可防止GLUT4mRNA表达量的降低 ,但罗格列酮对GLUT4mRNA表达量的降低无影响。结论 :低剂量STZ腹腔注射加高热量饲料喂养可导致大鼠类 2型糖尿病胰岛素抵抗样改变 ,益气活血通腑中药可改善此模型的糖耐量异常 ,其作用与增强其骨骼肌GLUT4基因表达 ,促进骨骼肌对葡萄糖的摄取和利用有关。 Objective:Effect of herbs for tonifying qi,activating blood circulation and clearing the hollow viscera (modified Taohechengqi decoction) and Metformin,Rosigilitazone on similar type 2 diabetic rats and the expression of glucose transporter 4 in their skeletal muscle were compared.Method:Similar type 2 diabetic with insulin resistance model were made by intraperitoneally administered low dosage of Streptozocin (STZ) in rats and fed with forage of high heat quantity.After the herbs,Metformin and Rosigilitazone were administered orally for 4 weeks in rats,the body weight were weighed,the sugar tolerance,serum insulin lipid in the blood and the expression of GLUT4 were determinated.Results:The body weight increased,the blood sugar significantly increased at the 30th,60th and 120th minute after intraperitoneally administered glucose,the fasting serum insulin did not change obviously,the HDL ch decreased in serum,mRNA expression of GLUT4 in the skeletal muscle reduced dramatically in diabetic rats.The body weight,fasting serum insulin and the 30th and 60th minute blood sugar after intraperitoneally administered glucose had no change in four drug groups.Low dosage herbs,Metformin and Rosigilitazone could reduce blood sugar at the 120th minute,but only Rosigilitazone could reduce the fasting blood sugar of this model.The herbs could prevent the lower of the serum HDL ch.Metformin could obviously insease the expression of GLUT4 in the skeletal muscle.The high dosage herbs could inhibit the decrease of the mRNA expression of GLUT4.Rosigilitazone had not any influence on the mRNA expression of GLUT4.Conclusion:Similar type 2 diabetic insulin resistant model were made by intraperitoneally administered low dosage of STZ and fed with the forage of high heat quantity.Herbs of Tonifying qi,activating blood circulation and clearing the hollow viscera could improve the impare glucose tolerance of this model.The action mechanism of the drugs may be related to promote the mRNA expression of GLUT4 in the skeletal muscle and improve the taking in and metabolism of blood glucose.
