热损伤对大鼠纹状体多巴胺受体基因表达的影响

摘要 目的 :研究热损伤对纹状体多巴胺受体D1R ,D2 R基因表达水平以及D2 R前体mRNA剪接的影响。 方法 :采用RT PCR的方法分析 4 3℃热损伤对大鼠纹状体多巴胺受体D1R ,D2 R基因表达的影响 ;用RNA酶保护实验检测 4 3℃热损伤后D2 RmRNA剪接的变化。结果 :4 3℃热损伤 30min后 ,D1R表达量显著降低 ,D2 R表达量升高。D2 R长短片段D2 RL/D2 RS的比值较对照组明显升高 ,D2 RL的转录占优势。 结论 :D1R ,D2 R及D2 R长短片段比值变化结果为阐明热损伤的分子机制提供了实验依据。 Objective: To investigate the effects of hyperthermia on the expression of D_ 1 R mRNA and D_ 2 R mRNA. Methods: RT-PCR was used to measure the expression of D_ 1 R mRNA and D_ 2 R mRNA in the striatum of rats with or without heat stress treatment at 43℃ for 30 min. Results: We observed that hyperthermia induced a decrease and increase in the expression of D_ 1 R mRNA and D_ 2 R mRNA respectively. The results of RNase protection assay indicate hyperthermia enhanced the ratio of D_ 2 RL mRNA/D_ 2 RS mRNA. Conclusion: It is suggested that heat stress may preferentially activate the transcription of D_ 2 RL.
