解毒祛瘀滋阴药并用激素对系统性红斑狼疮T细胞亚群Fas基因表达的干预作用研究

Study on the Adjustable Effect of Jiedu Quyu Ziyin Recipe on the Expression of Fas in T Lymphocyte Subsets in Systemic Lupus Erythematosus

目的 :研究解毒祛瘀滋阴中药并用激素对系统性红斑狼疮 (SLE)患者外周血T细胞亚群Fas表达的干预作用 ,探讨其免疫调节机理。方法 :5 3例女性SLE患者随机分为 2组 ,西药组以强的松治疗 ,中西结合组并用解毒祛瘀滋阴中药治疗。另选 30例健康女性作为正常对照。应用流式细胞仪检测各组治疗前后T细胞亚群 (CD3 + 、CD4+ 、CD8+ T细胞 )Fas基因表达的水平变化。结果 :治疗前与正常组比较 ,SLE组CD3 + 、CD4+ 、CD8+ T细胞Fas表达阳性率均明显升高 (均P <0 .0 1) ;活动期CD4+ Fas、CD8+ Fas表达阳性率均明显高于缓解期 (均P <0 .0 5 )。与本组治疗前比较 ,中西结合组治疗后CD3 + 、CD4+ 、CD8+ 细胞Fas表达阳性率均明显降低 (均P <0 .0 5 ) ;西药组CD3 + 、CD4+ 、CD8+ 细胞Fas表达阳性率也均明显降低 (均P <0 .0 5 )。两组间治疗后比较 ,CD8+ 细胞Fas表达率有统计学差异 (P <0 .0 5 )。结论 :解毒祛瘀滋阴中药与激素合用治疗SLE更能有效地调节T细胞亚群Fas的表达水平 ,促使机体紊乱的免疫内环境趋于平衡。

Objective:To investigate the adjustable effect of Jiedu Quyu Ziyin Recipe(JQZR) on the expression of Fas in T lymphocyte subsets of peripheral blood in systemic lupus erythematosus(SLE), and study its immunol regulation mechanism.Methods:53 female patients with SLE were divided into 2 groups randomly, integrated Chinese and Western medicine group being treated with JQZR plus prednisone and western medicine group with prednisone only.30 healthy female were chosen as nomal control group. Having measured the expression of Fas in T lymphocyte subsets (CD 3 +、CD 4 +、CD 8 +T cell) with flow cytometry (FCM).Results:Comparing with that in the normal group before treatment, all the expression of Fas in CD 3 +、CD 4 +、CD 8 + T cells in SLE group were higher obviously(all P<0.01);and the expression of Fas in CD 4 +、CD 8 + T cell in active stage were obviously higher than that in remission stage (P<0.05). Comparing with that in the same group before treatment, all the expression of Fas in CD 3 +、CD 4 +、CD 8 + T cells in the integrated Chinese and Western medicine group after treatment were decreased obviously, and so were those in the western medicine group. The expression of Fas in CD 8 + T cell in two groups after treatment were different significantly (P<0.05). Conclusion:JQZR together with prednisone can more effectively suppress the expression of Fas in T lymphocyte subsets in SLE patients and improve the disturbed immunol function.
