摘要 目的 通过雌激素对CCl4诱导的肝纤维化大鼠肝星状细胞活化及胶原合成影响的研究 ,探讨雌激素对肝纤维化的抑制作用。方法 CCl4诱导大鼠肝纤维化动物模型 ,观察雌激素 (苯甲酸雌二醇 1mg·kg-1)对大鼠肝纤维化形成过程中肝星状细胞的α 平滑肌肌动蛋白表达和对肝脏胶原含量的影响 ,并分别检测血清学标志及肝脏组织学等变化。结果 与肝纤维化模型组相比 ,雌激素可以明显降低大鼠血清ALT、AST水平 ,减少透明质酸和Ⅳ型胶原等纤维化指标的升高程度 (P均 <0 .0 5 ) ,抑制肝星状细胞对α 平滑肌肌动蛋白的表达和肝脏胶原沉积 ,减少肝脏I型胶原的合成。结论 雌激素可明显抑制肝纤维化大鼠肝星状细胞的活化及胶原合成 。 Objective To study the effects of estrogen on the activation of hepatic stellate cells and collagen synthesis in liver fibrotic rats, and then to investigate the suppressive effects of estrogen on liver fibrosis. Methods Liver fibrosis was induced in rats by CCl 4 administration, and was treated with benzoic estradiol (1 mg ·kg -1) twice weekly. The expression of hepatic stellate cells positive for a-smooth muscle actin (α-SMA), the collagen synthesis and other indicators were studied at the end of the experiment. Results Estradiol treatment reduced AST, ALT, hyaluronic acid (HA) and type Ⅳ collagen (CIV) in serums, suppressed hepatic collagen content, decreased areas of hepatic stellate cells positive for α-SMA, and lowered the synthesis of hepatic type I collagen significantly in the fibrotic rats induced by CCl 4 administration. Conclusion Estradiol suppresses the activation of hepatic stellate cells and collagen synthesis, thus reducing CCl 4-induced hepatic fibrosis in rats.
