摘要
建立了测定家兔血浆中灯盏花素主要有效成分灯盏乙素浓度的 RP- HPL C法 ,研究灯盏花素静脉注射给药后在家兔体内的药动学。色谱柱为 Diam onsil TMC18柱 (钻石 ,4 .6 m m× 2 5 0 m m,5 μm) ;流动相为 p H3.0的磷酸盐缓冲液 (0 .0 2 mol磷酸二氢钠用磷酸调 p H至 3.0 ,用 0 .4 5 μm水性滤膜滤过 ,即得。) -甲醇 -乙腈 (6 5 :35 :10 ) ;流速 1m l.m in- 1 ;柱温 2 5℃ ;检测波长 :334nm。灯盏花素静脉注射给药后主要有效成分灯盏乙素在家兔体内的药动学结果表明 ,灯盏乙素的药 -时曲线符合二室模型 ,主要药动学参数 t1 /2 α和 t1 /2 β分别为 1.2 9± 0 .5 3m in和10 .4 0± 1.97min;Vc为 14 8.1± 118.6 ml;CL 为 5 7.5± 31.7m l· min- 1。
An RP HPLC was established to determine scutellarin in rabbit plasma and to study pharmacokinetics of breviscapine in rabbits. The analytical column was DiamonsilTM C18(4.6 mm × 250 mm,5μm), the mobile phase consisted of methanol, 0.02 mol/L phosphate buffer( adjusted by phosphoric acid to pH 3.0) and acetonitrile (65∶35∶10),the flow rate was 1.0 ml·min 1 , the column temperature was 25 ℃ and the detection wavelength was 334 nm. The results of the pharmacokinetics study showed that the concentration time curve of scutellarin was comformed to two compartment model, the chief pharmacokinetic parameters were as follows: t 1/2 α 1.29±0.53 min, t 1/2 β 10.40±1.97 min, Vc148.1±118.6 ml, and CL 57.5±31.7 ml·min -1 .
