宁痫汤对戊四唑致痫大鼠脑内氨基酸类神经递质含量的影响 被引量:8

Effect of Ningxian Decoction on Cerebral Contents of Amino Acid Neurotransmitters in Rats with Pentylenetetrazol - Induced Epilepsy

摘要 【目的】探讨以升清降浊法组成的中药复方宁痫汤对实验性癫痫大鼠脑内氨基酸类神经递质含量的影响。【方法】以戊四哇(PTZ)诱导SD大鼠癫痫模型,观察中药复方宁痫汤(由升降散合温胆汤化裁组成)的抗痫效应,采用氨基酸分析法测定大鼠海马、皮层组织γ-氨基丁酸(GABA)、谷氨酸(Glu)的含量,并与正常组、模型组以及西药(苯巴比妥)组相对照。【结果】低剂量宁痫汤和西药苯巴比妥均能改善癫痫模型大鼠的惊厥行为(P<0.05),均能升高癫痫模型海马和皮层中GABA含量、同时降低其Glu含量(与模型组比较,P<0.05或P<0.01),并且两组的作用基本相仿(P>0.05),而高剂量宁痫汤的作用则比西药组差(P<0.05)。【结论】宁痫汤对癫痫发作有拮抗作用,其作用机制可能与调节脑内GABA、Glu水平,从而改善脑神经元兴奋--抑制平衡失调有关。 【Objective】To investigate the effect of Ningxian Decoction (ND), a complex prescription with the actions of ascending lucidity and descending turbidity, on cerebral contents of amino acid neurotransmitters in rats with epilepsy induced by pentylenetetrazol (PTZ) .【Methods】Forty rats were randomized into 5 groups; A (normal) , B (model) , C (high-dose ND) , D (low-dose ND) and E (positive control). PTZ-induced epilepsy rats were used to observe the antiepileptic effect of ND. Contents of gamma-aminobutyric acid (GABA) and glutamic acid (Glu) in hippocampal gyrus and cortex were detected by amino acid analysis. 【Results】Symptoms of convulsion was improved (P< 0.05) ; GABA content in hippocamal gyrus and cortex was increased and Glu content was decreased in Group D and E (Compared with Group B P<0.05 or P<0.01); the differences of GABA content and Glu content between Group D and group E were insignificant (P>0.05); the effect in group C was less than that in group E ( P<0.05). 【Conclusion】 ND can inhibit the occurrence of epilepsy and its mechanism may be relaxed to the regulation of cerebral contents of GABA and Glu.
