摘要
应用多种凝集素与6种上皮癌细胞株细胞、传代人羊膜胚胎细胞株细胞、正常人足月胚胎羊膜细胞和人A、B、O型血细胞、兔血细胞反应,检测其凝集素反应的程度;并通过荧光标记,测定凝集素受体在上述各型细胞膜上的分布;与此同时,还应用辣根过氧化物酶标记雪山豆凝集素,进行扫描电镜观察辣根过氧化物酶标记的雪山豆凝集素受体复合物在SGC肿瘤细胞表面上的分布情况.
The differences of agglutination between epithelioma cells (SGC,HSC,PLC, TCA, Hcla, Hep-2), FL cells and normal cells (normal mature amniotic cells, bloood type A, B, O cells and rabbit red cells) by twenty-three lectins were studied. In some reactions, the lectins were labeled by FTTC to study the binding of lectins to epithelioma cells. Results showed that SLL could strogly agglutinate epithelioma cells, but had no effect on normal cells; SML, CKL, PHA-s, APL, MDL could extensively agglutinate all kinds of epithelioma cells and norrnal cells; Lectins with blood group specificity such as BML, CML, PLA, MSL, MRL, UEL 1 could scarcely agglutinate epithelioma cells; The agglutination of the above cells by other lectins varied with the different types of lectin and cell. When SGC were treated with HRP-labeled SML, the HRP-SML-receptor complex could be observed by scanning electron microscope.
