摘要
以抗Bursin单抗(2F9-4/HU2),用免疫组化法,检测Bursin在鸭NH及1~12周龄不同免疫器官中定位分布,结果表明:NH期BF、Th、Sp及LN弱阳性,HG为阴性。1~12周龄内,各免疫器官阳性结果,变化不呈同步关系,其中BF以4周龄为典型,髓质内周缘阳性细胞层、FAE及FAESE为阳性、髓质中央阳性细胞稀少及IFSE中团簇状阳性细胞散在性分布,其余周龄期虽有相似情况,但相对阳性强度较弱。另外,12周龄时IFSE中仍有多量团簇状较强阳性细胞分布。Th以6、10、11周龄表现为强阳性细胞相对集中于髓质中类似于Hassall小体的区域,1周龄时阳性细胞较高密度弥散于髓质中,其余周龄阳性分布不典型。Sp以6、10、11周龄表现为强阳性细胞相对集中于髓质中类似于Hassall小体的区域,8周龄时阳性细胞较高密度弥散于髓质中,其余周龄阳性分布不典型。Sp以7~11周龄阳性细胞弥散分布于GC中,1~6周龄阳性细胞无特征性散在分布,12周龄时阳性细胞有呈团块状倾向。HG则1周龄为阴性,2~5周龄阳性细胞呈团块状散在于腺泡、腺管基底膜中,6~12周龄阳性细胞则无呈团块状表现。LN则1~2周龄弱阳性,3~12周龄强阳性细胞密布于淋巴小结中,除淋巴小结外,有的阳性细胞散在淋巴细胞聚集区或淋巴组织索中。
Stained with anti-Bursin monoclonal antibody (Mcab)2F9-4/HU2 by immunohi-stochemistry,the Bursin-containing immune organs and tissues were studied on new hatching (NH)and post-hatching duck,from 1 to 12 week-old.The results showed as following:There were weak positive effects of Bursin on bursa of Fabricius (BF),thymus,spleen and lymphoid node but the effect on harderian gland in NH Duck was negative. But the changes of positive effects of Bursin among different immune organs from 1 to 12 week-old ducks were not synchronous. In the 4 week-old duck, the inside fringe cells in medulla and the Follicle-Associated Epithelium (FAE) and FAESE of BF were typically positive while only a little positive in medulla central cell of BF and cluster positive cells were scattered in the Inter-Follicular Surface Epithelium(IFSE),but there was comparatively weak positive reaction at the other ages. In addition,cluster positive cells in IFSE were larger in 12 week-old duck.Strong positive cells in thymus collecting in the Hassall corpuscle-like area were found in medulla at 6,10,11 week-old days and mass positive cells were diffused in medulla at 8 week days; There was no distinct positive cells in thymus at other week days. In 7~11 week-old days in spleen positive cells diffused in GC but In 1~6 week-old days,the positive cell in spleen were scattered irregularly. In 12 week-old days,positive cell in the spleen tended to be conglomerate.In respect to HG,there were no positive cells in 1 week-old days while in 2~5 week-old days positive cells spread in the basement membrane of alveoli and gland duck as conglomeration but no cells spread as conglomeration in 6~12 week-old days.In LN ,positive cells were weak in 1~2 week-old days and the positive cells spread densely in lymphatic nodules at 3~12 week-old days. There were positive cells in the mass lymphoid cells area or in the lymphatic tissue cords except for the lymphatic nodules.
