摘要
研究了菜心种子、茎尖和愈伤组织离体筛选耐羟脯氨酸变异的方法。在多代连续选择中0.3mg/mL的羟脯氨酸对茎尖和愈伤组织是一个适宜的选择压力,经3~4个周期筛选后可获稳定入选株。种子萌发后的幼苗在1.0mg/mL浓度上经4个世代连续筛选可获抗性株系。3mg/mL浓度可用于幼苗的前期淘汰。入选系耐羟脯氨酸的能力和耐热性提高。
Techniques for screening in vitro of hydroxyproline-resistant lines were established with seedling, shoot and callus of Brassica campestris L. ssp.chinensis var.utilis Tsen et Lee, and some resistant lines were selected. An adaptable Hyp concentration is 0.3 mg/mL on which stable resistant lines from shoots and callus could be obtained by successive screening for 4 culture cycles. Resistant seedlings could be selected from the survivals cultured with 1.0 mg/mL Hyp for 4 cycles.One step-selection with above lethal dose was also tested. Selected lines showed high resistance to Hyp and the improvement of heat-tolerance.
