外源性孕酮对妊娠小鼠着床早期子宫内膜转化生长因子β_1表达的影响 被引量:2

The Effects of Progesterone on TGFβ_1 Protein Expression in Mouse Endometrium during Early Implantation

摘要 应用免疫组织化学 SABC法 ,研究了外源性孕酮对妊娠小鼠着床早期子宫内膜内转化生长因子 β1 (TGFβ1 )表达的影响。结果显示 :试验组皮下注射不同剂量孕酮 (2 mg/只 ,4 mg/只 )后 ,在孕 4 d,上皮内 TGFβ1 阳性着色 ,有的腺导管内具有 TGFβ1 阳性着色 ;且随着孕酮剂量的增大 ,上皮内 TGFβ1 阳性着色由子宫腔面向基面延伸 ,而腺上皮的阳性着色则呈下降趋势 ,基质内 TGFβ1 阳性着色则呈上升趋势。在孕 5 d,上皮内 TGFβ1 阳性着色呈现随孕酮剂量增大而下降的趋势 ,腺上皮和基质的 TGFβ1 动态变化如同孕 4 d。在孕 6 d,上皮、腺上皮及基质 TGFβ1 阳性着色均与孕 5 d的变化趋势相同。由此认为 ,外源性孕酮对小鼠子宫内膜各组织 TGFβ1 蛋白表达有影响。主要表现为 ,外源性孕酮可促进基质中 TGFβ1 的产生 ,对腺上皮 TGFβ1 The effect of s.c. administration of progesterone on the expression of TGFβ_1 in the mice endometrium during early implantation was studied by immunohistochemistry SABC technique.Results:in expermental groups(progesterone 2 mg/mouse and 4 mg/mouse),targets tissues are immunostained,but the pattern of changes is different from that in control groups:at day 4 pregnancy,TGFβ_1 protein expression in luminal is increasing with the increase of the dose of progesterone.Moreover,TGFβ_1 protein is extending from the side of the surface to the side of the stroma.;TGFβ_1 protein expression in glandual epithelial cells is decreasing and increasing in stromal cells with the increase of the dose of progesterone,and little TGFβ_1 protein is in gland.At day 5 pregnancy,the pattern of TGFβ_1 protein expression is increasing first and then decreasing,and the pattern in both glandual epithelial and stroma cells is the same as it at day 4 pregnancy.At day 6 pregnancy,the pattern of TGFβ_1 protein expression in target tissues is same as it at day 5 pregnancy.Conclusion:the s.c. administration of progesterone has the effects on TGFβ_1 protein expression in mouse endometrium:including:stimulate the production of TGFβ_1 protein and downregulate TGFβ_1 protein expression in glandual epithelial.
