As_2O_3注射液体外对人大肠癌细胞抑制作用的观察

In Vitro Inhibiting Effect of Arsenic Preparation on Cancerous Cells of Human Large Intestine

目的 明确中药砒霜制剂三氧化二砷 (As2 O3 )注射液对人大肠癌细胞的体外抑制作用 ,为临床应用As2 O3 治疗大肠癌提供实验依据。方法 采用活细胞观察法、台盘兰拒染法、四甲基偶氮唑蓝比色 (MTT)法和透射电镜观察了As2 O3 对体外培养的人大肠癌细胞株CoLo-3 2 0生长的影响及其形态学变化。结果 As2 O3 能显著地抑制人大肠癌细胞CoLo -3 2 0的生长 ,用药后诱导其出现了典型的细胞凋亡超微结构改变。结论 As2 O3 对人大肠癌细胞具有显著抑制生长作用 ,该抑制作用与诱导细胞凋亡密切相关。

OBJECTIVE To identify the inhibiting effect of the arsenic preparation (As 2O 3) on cancerous cells of human large intestine. METHOD Living cell observation method, trypan blue chromophobic method, MTT method and transmission electron microscopy were used to observe the effect of As 2O 3 on the growth and morphological changes of the in vitro cultivated human cancerous cell strain (CoLo - 320). RESULT As 2O 3 could markedly inhibit the growth of CoLo - 320, with typical changes in ultrastructure of apoptosis. CONCLUSION As 2O 3 has marked effect in inhibiting growth, which is closely related to the induction of apoptosis.
