摘要
目的 观察葛根素和灯盏花素对N -甲基 -N -亚硝脲 (N -methyl -N -ni trosourea ,MNU)诱导SD大鼠视网膜变性的保护作用及机制。方法 雌性SD大鼠随机分为正常对照组、模型组、葛根素和灯盏花素组。通过视网膜形态学分析测量周边视网膜的总视网膜厚度 ,TUNEL试剂盒检测光感受器细胞凋亡。结果 MNU处理后 7天模型组周边视网膜的总厚度是 3 6μm ,而葛根素组和灯盏花素组的周边视网膜总厚度分别是 46μm、 3 5 μm、 5 8μm、 43 μm和3 9μm。MNU处理 2 4小时后 ,模型组周边视网膜的光感受器细胞凋亡指数是 3 8 0± 3 6( % ) ,葛根素组和灯盏花素组的凋亡指数分别是 2 5 4± 3 0 ( % )、 3 9 0± 2 5 ( % )、 19 4± 1 9( % )、2 8 0± 3 0 ( % )和 3 6 9± 2 1( % )。结论 葛根素和灯盏花素对MNU引起的周边视网膜损伤有一定的保护作用 ,呈剂量依赖性 ,其作用机制是通过抑制光感受器细胞发生凋亡。
OBJECTIVE To observe the protective effect of puerarin and breviscapin on retinal degeneration induced with N-methyl-N-nitrosourea(MNU) in rats and its mechanism METHODS Female rats were randomly divided into control group, model group, puerarin groups, and breviscapin groups Retinal morphologic analysis was performed and the peripheral retinal thickness was measured, and apoptosis of photoreceptor cells was detected by TUNEL labeling RESULTS At 7 days total retinal thickness treated by MNU was 36μm, and these values were 46μm, 35μm, 58μm, 43μm and 39μm respectively in the puerarin groups and breviscapin groups At 24 hours apoptosis indexes of retina by MNU were 38 0±3 6(%) in the model group, and there were 25 4±3 0(%)、39 0±2 5(%)、19 4±1 9(%)、28 0±3 0(%)和36 9±2 1(%) respectively in the puerarin groups and breviscapin groups CONCLUSION The use of puerarin or breviscapin was effective in the suppression of MNU-induced peripheral retinal damage via anti-apoptotic mechanism and dose-dependent While they had no protective effect on MNU-induced central retinal damage
