摘要 目的 研究大鼠心脏移植排斥模型中外周血T淋巴细胞亚群表面CD15s、CD44表达,探讨其在急性排斥中的作用。方法 建立稳定的Ono氏大鼠腹部心脏移植急性排斥模型;应用流式细胞术观察外周血T淋巴细胞亚群比值及各亚群中CD15s、CD44的表达变化。结果 急性排斥时淋巴细胞CD15s在CD4+亚群中阳性率降低而荧光强度升高,CD8+亚群中CD15s荧光强度升高,CD44阳性率在CD8+亚群中降低而荧光强度在CD4+及CD8+亚群中均升高。结论 CD15s、CD44表达变化可反映同种异体移植后急性排斥状况,可能取代传统的膜表面分子,成为新的免疫粘附分子监测指标。 Objective To study the CD15s and CD44 expression and their roles in an acute rejection model of rat heart transplantation. Methods The Ono' s rat heart transplantation acute rejection model was established. The dynamic changes of CD15s,CD44 expression in PBLC of rat Ono's model were investigated using flow cytometry and every specimen of corresponding end points are performed pathologic study and there acute rejection degree are evaluated according to ISHLT standard. Results We found that CD4/CD8 increases whether rejection appears or not. While acute rejection, CD15s expression rates decreased in CD4 + subset and CD44 expression rates in CD8+ subsets,and their fluorescence inten-sities both increased in CD4+ and CD8+ subset. Conclusion CD15s and CD44 changes can reflect the acute rejection status after allotransplantation. They may be new immunological adhersion molecules to replace traditional membrane surface molecules as central monitoring roles.
