更昔洛韦对TK基因转染的淋巴瘤细胞的抑制作用

摘要 目的 探讨GCV对TK基因转染的淋巴瘤细胞生长的抑制作用。方法 利用缺陷型逆转录病毒介导的TK基因(RV-HSV-TK)转染淋巴瘤细胞,细胞增殖实验(MTT法),研究GCV对转染TK基因阳性淋巴瘤细胞株杀伤作用。结果 转染了TK基因的淋巴瘤细胞的生长曲线与未转染TK基因的细胞无差异。0.1~50μg/ml的GCV对转染阳性细胞有明显的毒性作用,杀伤效应与时间成正比。Hut-tk细胞对Hut细胞具有旁观者效应。结论RV-HSV-TK系统对淋巴瘤细胞有较高的转染效率,转染了TK基因的淋巴瘤细胞对GCV敏感。 Objective To study the inhibitory effect of ganciclovir(GCV) on lymphoma cells transfected with TK gene. Methods By using RV-HSV-TK gene system and MTT technique, the growth inhibiting effect of GCV on lymphoma cells tramsfected with TK gene was analysed . Results Herpes TK gene was successfully transmitted into lymphoma cell lines by retrovirus. GCV in concentrations ranging from 0.1 to 50ug/ml showed significant toxicity on lymphoma cells with TK gene; the effect was time dependent. By-slander effect was observed after coin-cubation of Hut-tk cells with Hut cells. Conclusions RV-HSV-TK system has high efficiency in transfecting lymphoma cells with TK gene by mediation of retrovirus,and these cells are sensitive to GCV.
