钴铬钼颗粒对大鼠骨髓基质干细胞cbfalmRNA的抑制

摘要 目的:探讨钴铬钼(Co-Cr-Mo)颗粒对大鼠骨髓基质干细胞(BMSC)成骨特异性转录因子cbfal(corebinding factor a l)mRNA的影响。方法:条件培养液(含0.00%、0.005%和0.001%的Co-Cr-Mo颗粒)干预第7d与第14d细胞,Annexin V法检测凋亡率、RT-PCR和原位杂交检测cbfalmRNA变化,结果:随着培养时间延长和颗粒浓度增加,BMSC凋亡率增加,cbfalmRNA表达减少,与对照组显著差异(P<0.05),cbfalmRNA阳性细胞数目降低,与对照组显著差异(P<0.05)。结论:Co-Cr-Mo颗粒引起BMSC的cbfalmRNA减少,降低其成骨分化能力。 Objective:To investigate the influence of Co-Cr-Mo particles to the expression of cbfalmRNA in rat BMSC(bone marrow stromal stem cells). Methods:The conditional media(containing three different concentration of Co-Cr-Mo particles:0. 00% ,0.005% ,0.01% )were applied to Rat BMSC having been cultured in 10% DMEM to 7-day-cells and 14-day-cells for 72 hours. The apoptosis rate was measured with Annexin V method, the expression of cbfalmRNA of BMSC was examined with RT-PCR and hybridization in situ. Results:When the conditional media applied to the 7-day-cells and 14-day-cells,apoptosis rate increased with the time and particles concentration increasing, the amount of cbfalmRNA and cbfalmRNA-positive cells gradually decrease with obvious difference with the cells in control groups(P<0.05).Conclusions:Co-Cr-Mo panicles can reduce the expression of CbfalmRNA in BMSC,and decrease the potential of osteogenic differentiation of BMSC.
