摘要
对 h EGF融合蛋白基因工程菌的发酵条件进行优化。结果表明 ,当起始 p H为 6 .6~ 7.0 ,以 10 0 m l/ 5 0 0 ml的装液量 ,6 %接种量 ,37℃培养 2 h,加入 0 .1mmol/ L的 IPTG诱导 4 h后 ,收获菌体可得到较高的生物量和 h EGF融合蛋白表达量。其中 IPTG诱导开始时间和菌体收获时间对蛋白表达有显著影响。
The fermentation conditions of recombinant fused hEGF were optimized. It was found that higher expression of fused hEGF could be achieved under the following conditions: 6% inoculum amount was added to 100 ml / 500 ml media at initial pH6.6 ~ pH7.0 . After 2 h of culture, 0.1 mmol/L IPTG was added to induce fused hEGF expression. The recombinant E coli . was harvested after 4 h of inducement. The results showed that the times of inducement and harvest were important factors for the expression of fused hEGF.
