摘要 目的 研究乳化异氟醚在大鼠静脉注射时的主要药效学参数。方法概率单位法测定乳化异氟醚和异丙酚在单次静脉注射时的半数有效量(ED_(50))及半数致死量(LD_(50)),并计算治疗指数(TI)。结果 静脉注射乳化异氟醚的ED_(50)和LD_(50)分别为:0.07和0.22 mL·kg^(-1),TI为3.0。静脉注射异丙酚的ED_(50)和LD_(50)分别为:5.89和18.19 mg·kg^(-1),TI为3.1。乳化异氟醚的麻醉恢复时间明显短于异丙酚(P<0.01)。结论 异氟醚乳化后静脉注射能产生确切的可逆性麻醉作用;乳化异氟醚的药效学安全范围与异丙酚相似;乳化异氟醚静脉给药后苏醒速度明显加快。 Objective To obtain the mainly pharmcodynamic data of intravenously emulsified isoflurane in rats, and to compare with those of propofol. Methods Median effective dose (ED50) and median lethal dose (LD50) were measured by probit analysis, and therapeutic index (TI) was calculated. Recovery times of anesthesia were compared between two agents by t-test. Results ED50, LD50(mL.kg-1)and TI of intravenously emulsified isoflurane were 0.07,0.22 and 3.00, respectively. For propofol, respective ED50,LD50 and TI were 5.89,18.19mg.kg-1 and 3.1. Recovery time of anesthesia for emulsified isoflurane was significantly shorter than that of propofol (P<0.01). Conclusion Induction and recovery of anesthesia is observed with intravenous injection of emulsified isoflurane. Recovery of anesthesia by intravenous use of emulsified isoflurane is more rapid than propofol, and this is the exactly value for new drug development.
