摘要
目的 :观察 c C1 DNA疫苗和蛋白质疫苗联合免疫对诱导 BAL B/ c小鼠免疫应答的影响。 方法 :雌性 BAL B/ c小鼠随机分为 4组 :A组 (DNA疫苗组 ) ,质粒 DNA以 1 0 d间隔免疫 3次 ;B组 (联合免疫组 ) ,质粒 DNA以 1 0 d间隔免疫 2次后以 GST- c C1融合蛋白加强免疫 1次 ;C组 (蛋白质疫苗组 ) ,分别在第 0、3周免疫接种融合蛋白 ;D组 (pc DNA3对照组 ) ,以 1 0d间隔 ,3次免疫接种质粒 pc DNA3。 EL ISA法检测血清样品中的特异性抗体水平以及实验小鼠脾脏淋巴细胞分泌 IFN- γ和IL- 4水平 ,以 MTT法行脾脏淋巴细胞增殖实验。 结果 :C组可诱导相对较强的小鼠免疫应答 ,其特异性 Ig G和 Ig G1以及脾脏淋巴细胞分泌 IL- 4水平均高于其他各组 (P<0 .0 5 ) ,免疫的类型以 Th2应答为主。B组诱导的特异性抗体水平和淋巴细胞分泌细胞因子水平均明显高于 A组 (P<0 .0 5 ) ,B组和 A组诱导的免疫应答类型均以 Th1应答为主。结论 :以 DNA prim ing-protein boost的策略可有效诱导较 DNA疫苗单独使用更高的特异性抗体应答 ,且以
Objective:To investigate the immune responses i nd uced by co immunizing with cC1 DNA vaccine and protein vaccine in mice. Methods: Female BALB/c mice were divided into 4 groups randomly. In gr oup A (DNA vaccine group), 3 successive immunizations with plasmid DNA were perf ormed at 10 d intervals. In group B (co immunization group), mice were primed w ith D NA vaccine and boosted twice with DNA and fusion protein respectively. In group C(protein vaccine group), each mouse was immunized with fusion protein GST cC1 at week 0 and week 3. In group D (pcDNA3 control group), mice were immunized with control plasmid 3 times at 10 d intervals.Specific antibodies in serum and cytok ines including IFN γ and IL 4 (secreted by splenocytes) were tested by ELISA. T h cell proliferation was determined by MTT method. Results: The mice in group C produced higher level of specific IgG, IgG1 and IL 4 (secreted by splenocytes) than those in other groups( P <0.05), and the main immune r es ponse was polarized to Th2 type. In group B, the level of specific antibody and cytokines were significantly higher than those in group A( P <0.05). The mai n immune response in group B and A were Th1 type. Concl usion: Compared with DNA vaccine immunization alone, the DNA prime prot e in boost vaccine combination can induce stronger specific immune respon se towards a Th1 type.
