摘要
目的 :测定商陆多糖的重均相对分子质量。 方法 :将商陆多糖分离纯化 ,采用 HPGFC- EL SD法测定重均相对分子质量分布。测定条件为 TSK柱 (G2 0 0 0 SW,7.5 mm× 30 0 mm) ;水为流动相 ,流速 1 .0 ml/ min;漂移管温度 4 0℃ ;气体 (N2 )压力 2 .0× 1 0 5Pa。结果 :多糖重均相对分子质量的线性范围为 1 0 0 0 0~ 1 70 0 0 0 (r=0 .973) ,测得商陆多糖重均相对分子质量约为 76 896 .4。 结论
Objective:To determine the distribution of weight averag e molecular weight in the Phytolacca acinosa Roxb. polysaccharide. Met hods : Phytolacca acinosa Roxb. polysaccharide was isolated and purified by high performance gel filtration chromatography (HPGFC) with evaporative light scattering detector (ELSD).Determination conditions were as fo llows: colum:TSK (G 2 000 SW 7.5 mm×300 mm);mobile phase:water; flow rate:1.0 ml/min;drif t tube temperature:40℃; nebulizer gas (N 2) pressure:2.0×10 5 Pa. Re sults: The linear range of weight average molecular weight of polysaccha ride was 10 000 170 000 ( r =-0.973). The weight average molecular weight of Phytolacca acinosa Roxb. polysaccharide was 76 896.4. Conclu sion: HPGFC ELSD is a practical, reliable method for the determination of the pol ysaccharide weight average molecular weight.
