摘要
目的 研究四环素对兔膝骨关节炎关节液中前列腺素 E2 (PGE2 )、一氧化氮 (NO)、一氧化氮合酶 (i NOS)以及大体形态学的影响 ,探讨其对骨关节炎防治的有效性。方法 36只日本大耳白兔随机分成正常组、对照组和治疗组。对照组和治疗组以 Hulth法造出骨关节炎模型。治疗组以四环素灌胃 ,正常组、对照组给予生理盐水 ;分别于 4w、8w抽取关节液 ,检测 PGE2 、NO、i NOS含量 ;对照组和治疗组进行关节软骨大体形态学的观察。结果 对照组关节液 PGE2 、NO、i NOS明显高于正常组 (P<0 .0 1 ) ;四环素能有效降低兔膝骨关节炎关节液 PGE2 、NO、i NOS的水平 ,4w分别降低 2 .8%、9.4%、1 2 .2 %;8w分别降低 2 .2 %、6.4%、7.8%;与对照组相比有显著性差异 (P<0 .0 5,P<0 .0 1 )。从大体观察 ,治疗组软骨状况明显优于对照组。结论 四环素对骨关节炎有一定保护作用。
Objective To explore the effects of tetracycline on the Prostaglandin E-2 (PEG-2), Nitric oxide (NO), Inducible NOS (iNOS) and gross morphometric change of cartilage in the model of experimental knee osteoarthritis (OA). And study the effect of tetracycline on the prevention and treatment of OA. Methods 36 Japanese rabbits were divided into normal group, control group and treatment group. The model of OA was established with Hulth technique in control group and treatment group. Rabbits in treatment group were administrated with tetracycline i.g., the control group andnormal group with NS. And the contents of iNOS, NO, PEG-2 in synovia were examined four weeks or eight weeks later. At the same time, the gross morphometric changes of cartilage were observed in control group and the treatment group. Results The expressions of iNOS, NO, PEG-2 in synovia, which have been effectively decreased 12.8%, 9.4%, 12.2% after 4 th week and 2.2%, 6.4%,7.8% 8 weeks after treatment respectively by tetracycline, of control group were much higher than the normal group remarkably ( P <0.01). It has remarkable difference between treatment group and control group ( P <0.05, P <0.01). The gross morphometric of cartilage in treatment group is superior to the control group. Conclusions Tetracycline has protective effect on OA.
