摘要
目的 探讨 p2 7kipl、CyclinD1在星形细胞肿瘤中的表达及意义。 方法 应用免疫组织化学方法检测p2 7kipl、CyclinD1在 4 5例星形细胞肿瘤组织及 10例正常大脑组织中的表达情况。结果 10例正常脑组织p2 7kipl阳性率为 80 .0 % (8/ 10 ) ,4 5例星形细胞肿瘤中 p2 7kipl阳性率为 35 .6 % (16 / 4 5 ) ;低度恶性星形细胞肿瘤(Ⅰ~Ⅱ级 ) p2 7kipl阳性率为 5 7.9% (11/ 19) ,高度恶性星形细胞肿瘤 (Ⅲ~Ⅳ级 ) p2 7kipl阳性率为 19.2 % (5 /2 6 ) ,差异有显著性意义 (P <0 .0 5 )。 4 5例星形细胞肿瘤CyclinD1阳性率为 4 2 .2 % (19/ 4 5 ) ,10例正常脑组织中CyclinD1无 1例阳性表达。p2 7kipl、CyclinD1表达均与星形细胞肿瘤的恶性程度有关 ,p2 7kipl表达与Cy clinD1表达呈显著负相关 (r =- 0 .775 ,P <0 .0 1)。结论 p2 7kipl蛋白表达的缺失可能与脑星形细胞肿瘤的发生、发展密切相关 ,p2 7kipl缺失与CyclinD1的过表达可能导致星形细胞肿瘤的发生。
Objective:To evaluate the significance of p27^(kipl) and CyclinD1 protein expressions and their relationship in the astrocytic tumors. Methods:The expressions of p27^(kipl) protein and CyclinD1 protein in 45 cases of astrocytic tumors and 10 cases of normal brain tissue were studied immunohistochemically. Results:Eight of 10 normal brain tissues were positive for p27^(kipl) protein. In contrast, the positive rate of p27^(kipl) protein in astrocytic tumor specimens was 35.6% (16/45) (P<0.01), the positive rate of p27 ^(kipl) protein in low grade astrocytic tumors (Ⅰ~Ⅱ ) was 57.9%(11/19) and 19.2% (5/26) in high grade astrocytic tumors(Ⅲ~Ⅳ) (P<0.05).The positive rate of CyclinD1 protein in astrocytic tumor specimens was 42.2%(19/45).None of the normal tissues exhibited CyclinD1 protein. The p27^(kipl) protein and CyclinD1 protein expressions were correlated with the malicious degree of the astrocytic tumors.The expression of p27^(kipl) was negatively related with the expression of CyclinD1(r=-0.775, P<0.01). Conclusion:The decrease of p27^(kipl) may play an important role in the genesis and development of astrocytic tumors,and may lead to tumor occurrence together with over-expression of cyclinD1.
