摘要
目的 探讨雷公藤多甙对人类红白血病细胞株K5 6 2作用及其机理。方法 应用四甲基偶氮唑蓝(MTT)法、荧光染色法、流式细胞仪 (FCM)等方法检测雷公藤多甙对人类红白血病细胞株K5 6 2的影响。结果 雷公藤多甙能显著抑制K5 6 2细胞的增殖 ,降低其存活率 ,其半数抑制浓度IC50 为 7.2 μg/ml;荧光显微镜下观察雷公藤多甙实验组的凋亡细胞增多 ;FCM检测雷公藤多甙实验组凋亡细胞百分率显著上升 (P <0 .0 5 )且G2 /M期细胞的百分率较对照组上升 (P <0 .0 5 )。结论 雷公藤多甙能显著抑制人类红白血病细胞株K5 6 2细胞的生长 ,促进细胞凋亡。
Objective: To study the mechanisms of effect on human K562 cells induced by the Glucosidorum Tripterygll Totorum. Methods:The Survival rate of cells were tested by MTT assay; Apoptotic cells were observed by fluorescence microscopy; The rate of sub-G1 phase cells and the change of the cell cycle were analyzed by flow cytometry.Results:Our results suggested that Glucosidorum Tripterygll Totorum could notably reduce the survival rate of K562 cells, IC 50 was 7.2?μg/ml;the rate of apoptosis cells and G2/M phase cells were increased in experimental group(P<0.05). Conclusions:Glucosidorum Tripterygll Totorum could notably inhibit the growth and proliferation of K562 cells and could induce the apoptosis of K562 cells.
