摘要
“纵现生命科学的发展趋势,最有代表意义的理论进展之一是越来越多地阐明细胞间、细胞内信号转导特征和机制。”细胞信号转导是生命科学迅速发展的前沿领域,自1991年~2000年间10个诺贝尔医学生理学奖项中,有6个奖项授与了细胞信号转导方面的工作。其概念已深入到生命科学的各个领域,成为解决许多理论和实践问题的基本思路。 本文将通过细胞信号转导知识的概括介绍,从配体(信号分子)与受体的结合,到G蛋白的转导,再到第二信使的产生,底物蛋白酶的生物效应及信号转导整合过程,剖析中药“多成分、多靶点、多途径、多效应”的现代科学内涵,以期实现中药疗效本质的分子水平解释,实现中药研究现代科学意义上的继承和发展,为中药作用机理的深入系统研究提供新思路、新方法。
Taking a panoramic view of the trends of development of life science, one of the most significant progresses in theory should be that in the characteristics and mechanisms of the transduction of intercellular and intracellular signals, which are being increasingly expounded. ' The transduction of cellular signals stands in the frontier area of life science, that is advancing with rapid strides. During 1991-2000, 6 of the 10 Nobel Prizes in medical science and physiology were rewarded to the research work in this field. The conception of the transduction of cellulcur signals has gone deeply into all the areas of life science and become a basic thinking in the resolution of quite a lot of theoretical and practical problems.
This article analyzes, by the way of a general introduction of knowledges in the transduction of cellular signals, the contents of modern science of the multi-components, multi-targets, multi-approaches and multi - effects'of the Chinese medicine from the combination of ligands (signal molecules) and receptors to the transduction of G protein and further to the generation of the second messenger as well as from the biological effects of substrate protease and the integration process of the transduction of signals, hoping to succeed in explaining the essence of the curative effect of the Chinese medicine at the cellular level and to realize the succession and development in the study of the Chinese medicine in the sense of modern science so as to provide the deepening and systematic study of the mechanisms of the effectiveness of the Chinese medicine with new thinking and methods.
