摘要
以硝普钠 ( Sodium Nitroprusside,SNP)作为 NO( Nitric oxide)的体外供体 ,以食管癌细胞系 EC1 0 9作为细胞模型 ,采用单细胞凝胶电泳法检测不同浓度的 NO作用不同时间后 ,食管癌细胞核 DNA被损伤的情况 .检测结果表明 ,在一定的条件下 ,NO对食管癌细胞核 DNA的损伤作用具有明显的浓度和作用时间依赖性特征 ,总的来讲 ,NO的浓度越大 ,NO对细胞作用的时间越长 ,细胞核 DNA所遭受的损伤越重 .
The injury of the nuclear DNA in esophageal carcinoma cell line EC109 as the cell model was examined by the single cell gel electrophoresis assay after exposed to sodium nitroprusside(SNP),the donor of NO in vitro,of different concentrations at various time.The results indicated that the damage effect of NO on nuclear DNA in esophageal carcinoma cells might depend on its concentration and its exposing time.The higher the concentration of NO is,the longer the time of NO exposing to esophageal carcinoma cells is,the more obvious the damage of nuclear DNA become under certain conditions.
