GDNF和HSV-GDNF对体外培养发育的大鼠骨骼肌卫星细胞Bcl-2表达的影响 被引量:4

摘要 为了进一步探索治疗神经系统损伤的新途径 ,本研究用传代培养的骨骼肌卫星细胞加入胶质细胞源性神经营养因子(GDNF )和单纯疱疹病毒介导的 GDNF(HSV-GDNF) ,取培养 2、4、8、16、2 4、48、72 h的细胞进行 Bcl-2单克隆抗体的免疫组织化学反应 ,观察了 GDNF和 HSV-GDNF对体外培养发育的大鼠骨骼肌卫星细胞 Bcl-2表达的影响。结果发现 :GDNF、HSV-GDNF组的肌卫星细胞 Bcl-2表达较对照组明显增强 ,细胞存活数量高于对照组 ,但 GDNF组、HSV-GDNF组两组之间无显著性差异。提示 :GDNF、HSV-GDNF可促进骨骼肌卫星细胞的增殖、分化 。 In order to investigate the effect of GDNF and HSV-GDNF on newborn rat satellite cells, myoblasts and myotubes of skeletal muscle ,we observed the expression of Bcl-2 on these cells in vitro. We established three groups (control, GDNF, HSV-GDNF group) after the third passage. The skeletal muscle cells were cultured for 2, 4, 8, 16, 24, 48, 72 h individually. Then these cells were stained with Bcl-2 monoclone immunohistochemical staining technique. The positive Bcl-2 immunoreactivity was present in the satellite cells, myoblast cells of skeletal muscle, but not in myotubes. It showed strongly in groups pretreated with GDNF and HSV-GDNF; and the number of the living cells is also more than that in the control group. The present results suggest that GDNF and HSV-GDNF might inhibit apoptosis, promote clonal expansion and differentiation on the cells of skeletal muscle mentioned above in virto.
