摘要
建立了人血清中卡铂的反相高效液相色谱测定法。固定相为Shim—packCLC—ODS柱,重蒸馏水作流动相,检测波长为229nm。卡铂平均绝对回收率为99.61%,日内和日间平均变异系数分别为4.18%和5.23%。最低检出浓度为0.05μg/ml,线性范围0.1~100μg/ml(r=0.9999)。应用本法测定了4例肿瘤患者静滴卡铂后的血药浓度。
A simple, rapid, accurate and precise reversed-phase HPLC method has been developed for the determination of carboplatin in human serum using Shim-pack CLC-ODS column (150mm×6mm ID). The mobile phase was distilled water and the eluent was monitored at 229 nm. Carboplatin elated at 6.1 min. The mean absolute recovery was 99.61%. Within day and clay-to-day precision (cv)were 4.18% and 5.23%, respectively. The measurable lowest level was 0.05ug/ml and the standard curve was linear over the concentration range of 0.1 100ug/ml (r=0.9999). The pharmacokinetic study of carboplatin in four cancerous patients was presented.
