摘要
目的:研究甘露糖结合凝集素(mannan-binding lectin,MBL)对高糖培养肾小球系膜细胞增殖和炎症因子分泌的影响。方法:体外培养系膜细胞,利用CCK-8法检测高糖和MBL对系膜细胞的增殖作用,并利用酶联免疫吸附法检测高糖和MBL对系膜细胞分泌相关炎症因子的影响。结果:高糖处理后,系膜细胞增殖能力增强,MBL可促进这一过程;高糖可促进系膜细胞分泌IL-6、IL-8、IL-17和TNF-β等炎症因子,MBL可加重系膜细胞这些炎症因子的分泌。结论:MBL能够加强高糖对系膜细胞增殖的促进作用,且能够促进高糖作用下炎症因子IL-6、IL-8、IL-17和TNF-β的分泌。
Objective:To investigate the effect of mannan-binding lectin(MBL)on the proliferation of mesangial cells and secretion of inflammatory factors under high glucose condition.Methods:The effects of MBL and high glucose on the proliferation of mesangial cells cultured in vitro were tested by CCK-8.And then the effect of MBL and high glucose on the secretion of related inflammatory factors were measured by ELISA.Results:After the treatment of high glucose,the proliferation activity of mesangial cells increased,as was promoted by MBL.High glucose could promote the secretion of inflammatory factors such as IL-6,IL-8,IL-17 and TNF-β,as could be enhanced by MBL.Conclusion:MBL can enhance the promotion effect of high glucose on the proliferation of the mesangial cells and can aggravate the secretion of IL-6,IL-8,IL-17 and TNF-βby the mesangial cells under high glucose condition.
