大豆异黄酮对骨肉瘤MG-63细胞的代谢和抗氧化应激研究 被引量:2

摘要 目的:观察大豆异黄酮(soybean isoflavone,SI)对人成骨肉瘤细胞株MG-63凋亡的诱导作用和线粒体抗氧化应激能力的改变。方法:不同浓度的SI作用于体外培养的MG-63细胞,采用蛋白印迹法检测免疫炎症细胞因子基质金属蛋白酶(metalloproteinase-2,MMP-2)和单核细胞趋化蛋白-1(monocyte chemotactic protein 1,MCP-1)表达,ELISA和分光度计法检测E-选择素(E-selectin)及高级氧化蛋白产物(advanced oxidation protein products,AOPPs)浓度变化,同时检测各组线粒体氧应激指标丙二醛(MDA)改变。结果:与正常对照组相比,SI各组患者MMP-2、MCP-1表达降低(P<0.05),E-selectin及AOPPs浓度降低(P<0.05),MDA含量增加(P<0.05);不同浓度SI之间的实验结果具有统计学意义。大豆异黄酮浓度为10.0 mg/ml组效果最显著,与2.5 mg/ml组及5.0 mg/ml组比较,差异有统计学意义(P<0.05)。结论:在MG-63的发生转化过程中,MMP-2、MCP-1、E-selectin、AOPPs及MDA可能起到重要作用,SI能够呈浓度依赖性抑制MG-63细胞增殖,诱导其凋亡,同时可增加MG-63线粒体抗氧化应激能力。 Objective:To investigate the apoptosis-inducing effect and anti-oxidative stress effect of soybean isoflavone(SI)on MG-63 cells.Methods:The cultured MG-63 cells were treated with different concerntrations of SI.Expressions of MMP-2 and MCP-1 were detected by western blot while concentrations of E-selectin and AOPPs were detected by ELISA and spectrophotometric methods.Results:Compared with control group,expressions of MMP2 and MCP-1 decreased(P <0.05),concentrations of E-selectin and AOPPs decreased(P <0.05)while concentration of MDA increased(P<0.05)in SI groups.There were significant differences between different concentrations of SI groups.The most significant effect was obseved in 10 mg/ml group(P<0.05).Conclusion:MMP-2,MCP-1,E-selectin,AOPPs and MDA may play an important role in the transformation of the MG-63 cells.SI can inhibit the proliferation and induce the apoptosis of MG-63 cell on a concentration dependent manner,and can increase the anti-oxidative stress ability of mitochondria in MG-63 cells.
