环孢素对人脐带间充质干细胞增殖、凋亡和细胞周期的影响

摘要 目的:探讨环孢素(CsA)对人脐带间充质干细胞(UC-MSC)增殖、凋亡和细胞周期的影响。方法:培养已被不同浓度CsA处理的UC-MSC,用MTT比色法观察各组UC-MSC增殖,用流式细胞术检测各组细胞凋亡及细胞周期。结果:CsA对UC-MSC增殖无明显抑制作用;CsA可促进UC-MSC凋亡,当CsA药物浓度≥240ng/ml,其细胞凋亡率与药物浓度呈正相关;CsA可改变UC-MSC细胞周期进程,使G0/G1期细胞比例下降,S期细胞比例增高,呈现浓度依赖性。结论:CsA对UC-MSC的细胞学功能无明显负作用。 Objective:To investigate the effects of CsA on the proliferation,apoptosis,and cell cycle of human umbilical cord mesenchymal stem cells(UC-MSC).Methods:UC-MSC were treated with CsA at different concentrations.The inhibitory effect of CsA on cell proliferation was detected by MTT assay,the apoptosis and cell cycle were detected by flow cytometry.Results:CsA had no inhibitory effect on cell proliferation of UC- MSC,but could induce apoptosis of UC- MSC.There was a increase trend of apoptosis rate when the CsA concentration was beyond 240ng/ml.CsA could also change cycle process of UC-MSC in concentration dependent manner,which could decrease cell proportion in G0/G1 phase and increase it in S phase.Conclusion:CsA has no obvious negative effect on the cell fuction of UC-MSC.
