大孔树脂纯化酸石榴汁中花青素的研究 被引量:7

摘要 【目的】筛选分离纯化酸石榴汁花青素的最佳树脂,优化酸石榴汁纯化的工艺条件,为石榴汁花青素的工业化生产提供参考。【方法】从AB-8、X-5、D101、D101A和NKA-9等5种大孔树脂中,通过静态吸附-解吸试验,筛选适合酸石榴汁花青素纯化的大孔树脂,分析花青素质量浓度、温度、pH及解吸液乙醇体积分数和pH对树脂静态吸附-解吸的影响,并在静态试验的基础上,通过动态吸附-解吸试验,确定最佳的吸附流速和解吸流速。【结果】AB-8树脂是纯化酸石榴汁花青素的最佳树脂,其对酸石榴汁花青素静态吸附-解吸的最优工艺条件为:室温25℃、pH 2.5、花青素质量浓度0.131 2mg/mL,解吸液采用体积分数70%酸化乙醇(pH 2.0);动态吸附-解吸的最适工艺条件为:吸附流速1.5mL/min,解吸流速2.0mL/min。【结论】AB-8大孔树脂对酸石榴汁花青素的纯化效果最佳,适用于酸石榴汁中花青素类物质的纯化。 【Objective】This study was conducted to screen the best resin for separation and purification of anthocyanins from pomegranate juice,and to determine the optimum purification conditions.【Method】Static adsorption and desorption analytical test was conducted to screen the best macroporous resin from AB-8,X-5,D101,D101 A,and NKA-9for purification of anthocyanins from pomegranate juice.Effects of the sample liquid concentration,temperature,pH,and concentration and pH of eluent were investigated and the optimal adsorption velocity and desorption velocity were obtained through dynamic adsorption-desorption test.【Result】AB-8resin was the most effective resin for adsorption and desorption of anthocyanins from pomegranate juice.The optimal conditions were temperature 25 ℃,pH 2.5,concentration of anthocyanin0.131 2mg/mL,and desorption agent 70%ethanol(pH 2.0).For dynamic adsorption-desorption,the best adsorption rate was 1.5mL/min and the best desorption elution flow rate was 2.0mL/min.【Conclusion】AB-8resin was the most effective resin for purification of anthocyanins from pomegranate juice.
