摘要
【目的】研究雌性山羊星状神经节中雌激素受体(Estrogen receptor,ER)的分布特点,探讨雌性山羊星状神经节神经元对雌激素是否具有反应性。【方法】取雌性山羊星状神经节,制作石蜡切片,经免疫组化SP法染色后,在数码显微镜下拍照,用高清晰图像分析系统对ER的分布特点进行分析,并计算相对表达量。【结果】在星状神经节的神经元胞体、卫星细胞和过路纤维中均有ERα、ERβ免疫阳性产物分布,但主要分布于神经元胞体。在神经元胞体中,细胞膜和细胞质为强阳性染色,而细胞核为弱阳性染色。ERα在神经元胞体中的相对表达量显著高于非神经细胞结构(P<0.05),ERβ在神经元胞体中的相对表达量极显著高于非神经细胞结构(P<0.01)。【结论】雌性山羊星状神经节神经元具有对雌激素刺激反应的受体,提示星状神经节可能作为雌激素作用于心血管的内分泌途径和自主神经对心血管调节的神经途径之间相互协调的网络节点,从而对心血管系统起调节作用。
【Objective】The purpose of this research was to study the localization of estrogen receptors(ER)on stellate ganglion of female goats and to explore whether neurons in stellate ganglion had reaction response to estrogen in female goats.【Method】Stellate ganglions were taken from mature female goats to prepare paraffin sections.The distribution characteristics of estrogen receptorsαandβon the stellate ganglion of female goats were then observed under digital microscope after immunohistochemical SP stain.【Result】ERαand ERβwere widely distributed in the stellate ganglion,and ERαand ERβimmunoreactive substances were distributed in neurons,supporting cells,and passing fiber.ERαand ERβwere mainly distributed in neurons.In neurons,ERαand ERβwere mainly distributed in cell membrane and cytoplasm,and the nuclear was weak positive.The relative expression level of ERαwas significantly higher than that of non-neurons(P<0.05),and ERβwas very significantly higher than that of non-neurons(P<0.01).【Conclusion】Neurons with receptor had reaction response to estrogen in stellate ganglion of female goats.The stellate ganglion was the network node that estrogen regulated cardiovascular system via the endocrine pathway and autonomic nerve pathway.
