生黄液对大鼠骨髓间质干细胞分化为神经细胞的定向诱导作用 被引量:2

摘要 目的探讨生黄醇提取液对成年大鼠骨髓间质干细胞 (MSCS)的体外增殖和特异性诱导、分化为神经细胞的能力。方法采用生黄醇提取液、2 — 硫基乙醇 ( 2 — ME)、二甲基亚枫 +成纤维细胞生长因子 (DMSO+FGF)、1 640培养液 +1 5 %FeS(对照 ) 4组体外定向诱导MSCS 向神经元细胞分化 ,用倒置显微镜和免疫组化技术检测神经元细胞所表达的特异性标志。结果 3种不同诱导液均能定向诱导MSCS 分化为神经元细胞 ,其中 ,生黄液组的NSE(神经元特异性烯醇化酶 )和GFAP(胶质纤维酸性蛋白 )的阳性细胞数明显高于 2 —ME和DMSO +FGF组 (P <0 .0 5 ) ,1 640培养液 +1 5 %FeS组没有诱导作用。结论MSCS 能在体外扩增、传代、能被定向诱导分化为神经元细胞。 Objective To explore the potential ability of Shenghuang Extract Solution(SES) to induce oriented differentiation effect on the adult rat mesenchym stem cells(MSC S)into neuron-like cells(NLC S) and the ability of MSC S to proliferate in vitro. Method MSC S were orientedly induced to differentiate into NLC S in vitro, cultured in the media added with SES, 2-ME and DMSO plus FGF respectively and NLC S were identified by immunohistochemical techniques via determing the expression of specific markers on them.Results All the three kinds of inducing substance could induce oriented differentiation of MCS S into NLC S.However,the positive cell number with the expression of neuron-like enolase(NSE) and glial fibrillary acidic protein(GFAP) were significantly higher in the group of cultures with SES than those with 2-ME and DMSO plus FGF(P<0.05). Conclusions MSC S can proliferate and be passed on from generation to generation.Especially,they can be induced orientedly to differentiate into NLC S.
