摘要
目的 研究昆布多糖硫酸酯 (LAMS)对急性髓性白血病HL - 6 0细胞株NF -κB表达的影响及CD11b在HL - 6 0细胞株的表达。方法 用MTT法测定不同浓度LAMS对HL - 6 0细胞株的增殖抑制作用 ;流式细胞仪分析不同浓度作用下细胞凋亡率 ,免疫组化法测定MMS对HL - 6 0细胞株NP -κB家族P6 5亚单位的作用及CD11b表达。结果 HL - 6 0细胞增殖抑制程度与昆布多糖硫酸酯的浓度呈正相关 ;HL - 6 0细胞凋亡率随药物浓度的增加而升高 ;在LAMS浓度为 0 .75 μg/ml作用下 ,P6 5在 12h表达呈强阳性 ,2 4h几乎不表达 ,而IKbα在 6h表达呈强阳性 ,CD11b不表达。结论 LAMS可以诱导体外HL - 6 0细胞凋亡 ,其中对凋亡的调控作用可能为负调控 ;LAMS能够调控NF -κB的表达 ,其表达有时间差异性。说明LAMS已作用于分子水平 ,且对CD11b在HL - 6 0细胞株的表达无影响。
Objective: To study the effects of LAMS on modulation of HL-60 cell strain and to study expression of CD11b on ITL-60 cell strain. Methods:MTT was used to determine the inhibiting action in the different concentration of LAMS on HL-60 cell strain; The attitude of Sub-G1 peak in DNA histogram was examined by FCM; NF-κB expression affected by lAMS was determined by using immunohistochemical stainings. Results: With the increase of LAMS concentration, the inhibiting effects on multiplication of HL-60cells were decreased ; After treatment of HL-60 cells with 0.75?μg/ml LAMS for 6?h, 12?h , 24?h, expression of P65 was the most significant at 12?h, while the expression of IKba was the most significant at 6?h. The expression of NF-κ B reduced in LAMS group without time-dependent pattern. CD 11b do not express.Conclusions: LAMS can induce HL-60 cells opoptosis. LAMS can modulate the expression of NF-κB . It also suggested that LAMS bad an influnced at molecul level. LAMS do not affect on the expession of CD11b on HL-60 .
