新疆维、汉族妇女宫颈病变中p14^(ARF)表达及其与HPV感染的关系

The relationship of p14^(ARF) expression and HPV infection in cervical lesions between Uygur and Han Women in Xinjiang

摘要 目的:探讨p14ARF在新疆维、汉族妇女宫颈病变的表达情况及其与HPV感染的关系。方法:根据病理诊断结果,将维、汉族妇女宫颈病变组织各120例,分为慢性炎症组、CINⅠ组、CINⅡ~Ⅲ组及宫颈鳞癌组,每组各30例。免疫组化SP法检测p14ARF表达,PCR-反向点杂交法检测HPV分型。结果:不同级别宫颈病变组织中p14ARF表达差异显著(P<0.05),且随着病变程度加重而增加。p14ARF在宫颈病变组织中的表达无民族差异(P>0.05)。维、汉族宫颈病变的HPV阳性率无显著差异(51.7%vs 54.2%,P>0.05)。随着宫颈病变程度的加重,HPV阳性率增高(P<0.05)。HPV阳性宫颈病变组织中p14ARF蛋白阳性表达率显著高于HPV阴性病变组织(P<0.05),且p14ARF蛋白表达与HPV感染呈正相关(r=0.480,P<0.05)。结论:p14ARF蛋白在宫颈病变及癌变中的表达异常可能与HPV感染有关;p14ARF在宫颈病变组织中的表达在民族间无差异。 Objective: To explore p14ARFexpression in cervical lesions between Uygur and Han women and its relationship with HPV infection in Xinjiang. Methods: 120 Uygur women and 120 Han women with cervical lesions were retropectively studied from Mar. 2009 to May. 2013 who at Obstetnics and Gynecology department Urumuqi General Hospital of Lanzhou Military Region. Specimens were divided into four groups,including chronic cervicitis group, CINⅠ group,CINⅡ ~ Ⅲ group and cervical squamous cell carcinoma group,30 women in each group. Immunohistochemistry SP method was used to detect the p14ARFexpression. PCR-RDB method was applied to detect HPV genotypes. Results: With the deterioration of cervical lesions,the expression of p14ARFprotein had significant differences between different groups( P < 0. 05) including chronic cervicitis group and CIN Ⅰ group,CIN Ⅱ ~ Ⅲ group and cervical squamous cell carcinoma group. The differences of p14ARFexpression between Uygur and Han nationality was significant( P > 0. 05). HPV positive rate of Uygur and Han nationality in cervical lesions were 51. 7 % and 54. 2% respectively. There was no significant difference of HPV positive rate in the two nationalities( P > 0. 05),while with the deterioration of cervical lesions degree,HPV positive rate had notable rise( P < 0. 05). p14ARFprotein expression in HPV positive groups was obviously higher than that in HPV negative group( P < 0. 05),and its high expression had correlation with HPV infection( r = 0. 480,P < 0. 05). Conclusion: Abnormal ex-pression of p14ARFand HPV infection are closely related to development of cervical lesions and cervical cancer. p14ARFprotein expression has no significant difference between Uygur and Han nationality.
