摘要
目的:探讨上皮性卵巢癌组织中胰岛素样生长因子ⅡmRNA结合蛋白1(IMP1)蛋白及mRNA的表达与上皮性卵巢癌发生发展的关系。方法:采用免疫组化法和RT-PCR技术检测47例上皮性卵巢癌(恶性组)、28例上皮性卵巢良性肿瘤(良性组)及20例正常卵巢上皮组织(正常组)中IMP1蛋白和mRNA的表达情况,并分析IMP1蛋白表达与上皮性卵巢癌的临床病理特征的相关性。结果:上皮性卵巢癌、上皮性卵巢良性肿瘤及正常卵巢上皮组织中IMP1蛋白的阳性表达率分别为85.1%、32.1%和10%,IMP1mRNA相对表达量分别为0.652±0.067、0.120±0.014和0.085±0.011。卵巢癌组织中IMP1蛋白及mRNA表达量均显著高于正常组和良性组(P<0.05),而后两组则无显著差异(P>0.05)。上皮性卵巢癌组织中IMP1蛋白表达与FIGO分期和病理组织学分级有关(P<0.05),而与有无腹水、淋巴结转移无关(P>0.05)。结论:IMP1可能参与上皮性卵巢癌的发生发展过程。
Objective: To investigate the expression of insulin-like growth factorⅡ mRNA-binding protein1( IMP1) proteins in epithelial ovarian cancer tissues as well as its mRNA, and the relationship with the development of ovarian cancer. Methods: The expression of IMP1 proteins and mRNA in 47 specimens of epithelial ovarian cancer,28 specimens of epithelial benign ovarian tumor and 20 specimens of normal ovarian epithelial tissues were measured by immunohistochemistry and reverse transcription-polymerase chain reaction respectively. The correlations of the expression of IMP1 proteins to the clinicopathologic characteristics of epithelial ovarian cancer were analyzed. Results: The positivity rate of IMP1 proteins were 85. 1%,32. 1% and 10% in epithelial ovarian cancer,epithelial benign ovarian tumor and normal ovarian epithelial tissues. The levels of IMP1 mRNA expression were 0. 652 ± 0. 067,0. 120 ± 0. 014 and 0. 085 ± 0. 011,respectively. The expression of IMP1 proteins and mRNA were significantly higher in epithelial ovarian cancer than those in epithelial benign ovarian tumor and normal ovarian epithelial tissues( P < 0. 05). The expression of IMP1 protein was correlation with surgery clinical stage and histologic grading,while no correlation with peritoneal lymph node metastasis and ascites. Conclusion: IMP1 may participate in the development of epithelial ovarian cancer.
