基于近红外光谱法快速检测裸花紫珠颗粒中木犀草苷和毛蕊花糖苷的含量

Rapid Determination of Luteolosided and Verbascoside in Callicarpa Nudiflora Granule Based on Near-infrared Spectroscopy

目的采用近红外光谱法建立裸花紫珠颗粒中木犀草苷和毛蕊花糖苷含量的快速检测方法。方法采用HPLC测定裸花紫珠颗粒中木犀草苷和毛蕊花糖苷的含量,利用近红外光谱法结合偏最小二乘法(PLS),建立裸花紫珠颗粒中木犀草苷和毛蕊花糖苷的定量校正模型。结果木犀草苷和毛蕊花糖苷的定量校正模型的相关系数(r)分别为0.987 9,0.956 3;内部交叉验证均方差(RMSECV)分别为0.004 6,0.063 7。验证集的相关系数(r)分别为0.905 8,0.964 8;预测误差均方差(RMSEP)分别为0.020 0,0.079 8。结论该研究基于近红外光谱法建立的裸花紫珠颗粒木犀草苷和毛蕊花糖苷快速检测定量模型性能良好,预测结果良好,为中药颗粒的含量测定和质量控制研究提供了科学合理的方法。

OBJECTIVE To establish a rapid detection method to detect the content of luteolosided and verbascoside in Callicarpa Nudiflora granule by near infrared spectrum.METHODS Determined the content of luteolosided and verbascoside in Callicarpa Nudiflora granule by HPLC.And used near infrared spectroscopy combining with partial least squares(PLS)to set up quantitative calibration model of luteolosided and verbascoside.RESULTS The correlation coefficients(r)of the quantitative correction model of luteolosided and verbascoside were 0.987 9 and 0.956 3 respectively.The values of root mean square error of cross validation(RMSECV)were 0.004 6 and 0.063 7.The correlation coefficient(r)of the verification set were0.905 8 and 0.964 8 respectively.The mean square error(RMSEP)were 0.020 0 and 0.079 8.CONCLUSION Based on near infrared spectroscopy,the study establish a rapid quantitative model of luteolosided and verbascoside and its performance is good,and the prediction results of the model are good.This study provides a scientific and reasonable method for the content determination and quality control of Chinese medicine granules.
