摘要
急性心肌梗塞大鼠,给予异搏定(0.8mg/100g,ip)后,与对照组相比,显著提高了对容量负荷的耐受力;且梗塞范围明显缩小。慢性心肌梗塞大鼠,给予异搏定(0.8mg/100g,ip)后,与对照组相比,显著缩小了梗塞范围和代偿性肥大程度。异搏定缓解心绞痛有效,且对急性缺血的心肌有保护作用,已被证明。但其对CMl的影响,尚未见报道。本实验证实了异搏定对AMl大鼠有治疗作用,并探讨了其对大鼠CMI病程的影响。
This study was conducted to confirm the effect of verapamii (V) upon acute myocardium infarction (AMI) in intact anesthetized rat model and explore its long term efficacy in chronic model (CMI) . In AMI group, 13 rats received one dose infusion of V (0.8mg/100g ip) 1 hour prior to coronary occlusion, and 13 received normal saline infusion. At rest, the hemodynamic parameters were not significantly different between two groups. While the work load of the heart was increased by rapid administration of saline (0.4ml/ 100g, i. v.), left ventricular systolic pressure and diastolic pressure were decresed6.5±1.8% (P<0.01)and 24±9.3% (P<0.05)(which were found to be less in V-treated group respctively) and restored to base line significantly faster than those of controls. In addition, infarct size was reduced by 15.1±4% (P<0.01). There were no significant differences in hemodynamic indices, scar thickness and the maximal diameter of the left ventricular cavity between two groups. However, in V-treated group infarct size was 5.3±1 .7 % (P<0.02) smaller and compensatory hypertrophy was relatively less compared to controls,
