结核杆菌感染对人肺组织β-防御素的作用 被引量:4

摘要 目的 探讨结核杆菌对人肺组织 β -防御素 -1,2基因的表达调控。 方法 取肺结核病人病变部分的肺组织 ,提取RNA ,采用RT -PCR技术扩增人 β -防御素hBD -1cDNA ,hBD -2cDNA ,并重组到pGEM -TEasy载体 ,经限制性内切酶谱分析和DNA序列测定证实。结果 所克隆的cDNA片段分别为hBD -1,hBD -2的cDNA序列 ;肺结核病人肺组织hBD -诱导表达 ,hBD -1固有表达增强。结论 结核杆菌感染可正相调节人肺组织hBD -1,hBD ObjectiveTo study the regulation of human β-defensin-1, 2 gene expression by mycobacterium infection. MethodsRNA was isolated from mycobacterium- infected hum an lung tissue.The human β-defensin-1,2 cDNA fragment was obtained by RT-PCR am plification.The RT-PCR products around 300?bp,270?bp in size were cloned separ ately into pGEM-T easy vector and the recombinant clones were analyzed by digest ion with restriction endonucleases and DNA sequencing.ResultsSequence analysis confirmed that the RT-PCR produ cts were hBD-1 cDNA and hBD-2 cDNA respectively.The expression of hBD-2 gene was induced in mycobacterium-infected human lung tissue,while the expression of hBD -lgene was enhanced.ConclusionMycobacterium infection could up-regulate hBD- 1,2 gene expression in human lung tissues.
