摘要 目的 探讨大豆异黄酮对成骨细胞胰岛素样生长因子 -1(IGF -1)表达的影响。方法 体外培养新生大鼠颅骨成骨细胞 ,然后用塞唑盐 (MTT)法测定大豆异黄酮对成骨细胞的增殖作用 ,过氧化物酶标记的链霉卵白素(SP)免疫组织化学染色试剂盒检测IGF -1抗体表达 ,用原位杂交方法检测IGF -1mRNA基因表达 ,用HPIAS -10 0 0图象分析仪对各组表达进行定量分析。结果 大豆异黄酮培养的大鼠第 3代成骨细胞的IGF -1抗体和IGF -1mRNA呈阳性表达 ,并显著高于阴性对照组 (P <0 0 5) ,阳性细胞胞浆呈棕黄色阳性表达。结论 大豆异黄酮可通过促进大鼠成骨细胞IGF -1分泌而影响成骨细胞形成。 ObjectiveTo study the effect of soybean isoflavone on IG F-1 expression of osteoblastic cells. MethodsThe osteoblstic cells from cranial bone of newbor n rat was cultured.The effect of soybean isoflavone on osteoblastic cells prolif eration was measured by MTT.The Streptavidin-Peroxidase Histostain TM -Plus Kits was used to detect IGF-1 expression.ICF-1mRNA was detected with situ hybri dization.The quantitative analysis was monitored with HPIAS-1000 photograph anal ysis meter. ResultsThe IGF-1 and IGF-1 mRNA expression of the third generation osteoblastic cells cultured with soybean isoflavone was positive expr ession and was higher signifiantly than negative control group (P<0.05).The plasm of positive cells was brown and yellow expression. ConclusionThe soybean isoflvone may affect the formatio n of osteoblastic cells through promoting the secretion of IGF-1.
