miR-124-3p.1靶向PRDM2调节JAK/STAT通路对胰腺癌细胞增殖和凋亡的影响

Effect of mi R-124-3p.1 Targeting PRDM2 on Cell Proliferation and Apoptosis in PANC-1 via JAK/STAT Pathway

[目的]研究miR-124-3p.1对胰腺癌细胞增殖、凋亡的影响,并探讨其作用机制。[方法]运用q RT-PCR法检测组织和细胞中miR-124-3p.1、PR结构域蛋白2(PRDM2)的表达;将mi R-124-3p.1mimics、miR-NC、anti-miR-124-3p.1 mimics、anti-miR-NC用脂质体法转染PANC-1细胞;Western blot检测细胞中PRDM2、p-JAK2、p-STAT3蛋白表达;MTT法检测细胞增殖。[结果]与人正常胰腺组织细胞相比,胰腺癌组织细胞中PRDM2表达显著性升高,miR-124-3p.1表达显著性降低(P<0.05);PRDM2为mi R-124-3p.1的靶标。过表达miR-124-3p.1可抑制胰腺癌细胞增殖、促进凋亡、抑制JAK/STAT通路。[结论] miR-124-3p.1可抑制胰腺癌细胞增殖和促进凋亡,可能与靶向PRDM2和抑制JAK/STAT通路有关。

[Objective]To study the effect of miR-124-3 p.1 on cell proliferation and apoptosis of pancreatic cancer cells,and to explore its mechanism.[Methods]qRT-PCR was used to detect the expression of mi R-124-3 p.1 and PRDM2 in tissues and cells.miR-124-3 p.1 mimics,mi R-NC,anti-mi R-124-3 p.1 mimics,anti-miR-NC was transfected into PANC-1 cells by liposome method.The protein expression of PRDM2,p-JAK2 and p-STAT3 was detected by Western blot,cell proliferation was detected by MTT assay.[Results]Compared with normal pancreatic tissue and cells,the expression of PRDM2 was significantly increased,and the expression of miR-124-3 p.1 was significantly decreased in pancreatic cancer tissues and cells(P<0.05).PRDM2 was a target of miR-124-3 p.1.Overexpression of miR-124-3 p.1 inhibited pancreatic cancer cell proliferation,promotes apoptosis,and inhibited JAK/STAT pathway.[Conclusion]mi R-124-3 p.1 could inhibit the proliferation and promote apoptosis of pancreatic cancer cells,which may be related to targeting PRDM2 and inhibiting JAK/STAT pathway.
