TNF-α和CTGF在小鼠皮肤损伤修复过程中的表达及与损伤时间的关系 被引量:1

Experimental Study on Relationships Between TNF-α and CTGF Expression and Wound Age During the Healing Process of Wound on Mouse Skin

摘要 观察增生因子在小鼠皮肤切创组织不同时间的基因表达变化及其与损伤时间的关系。用免疫组织化学染色法和RT-PCR法,检测小鼠皮肤切创组织中不同时间的肿瘤坏死因子-α、结缔组织增殖因子基因表达的变化。皮肤切创后,在早期各基因表达水平较低,变化不明显,24h后开始上升,第6d上升至最高峰,两种生物因子的表达水平与损伤时间呈非直线性相关关系。损伤皮肤组织中两种被检因子的基因表达随伤后不同时间呈现一定规律性的非直线性相关变化,不适用于损伤时间的推断,但对同一组织多种细胞因子同时检查推测损伤时间是可行的。 The relationship between the changes of genetic expression of proliferative factor and wound age during different phases of incised wound on mouse skin was explored.The cytokines include TNF-α and CTGF were detected by using immunochemical staining and reverse transcriptase polymerase chain reaction(RT-PCR) at different phases after incised wound.Results showed that all genetic expressing levels were very low at early phase after injury and change was not obvious.They began to rise after 24 h and peaks were found at 6d after wound.Those two genetic expressing levels and injury time are nonlinearly related.The genetic expression of those two tested biological factors shows regular nonlinearly related change at different phase during the incised wound healing,which can't be used to estimate the wound age,but it works when multiple cytokines of the same tissue estimate the wound age at the same time.
