通痹灵、雷公藤多苷及青藤碱对CIA大鼠滑膜细胞凋亡与P_(53)表达的比较研究 被引量:12

摘要 目的 :研究通痹灵、雷公藤多苷及青藤碱对Ⅱ型胶原蛋白诱导性大鼠关节炎模型 (CIA)滑膜组织细胞凋亡及P53蛋白表达的影响。方法 :原位细胞凋亡检测试剂盒检测各组原代大鼠滑膜细胞凋亡的水平 ,免疫组化法检测各组原代大鼠滑膜细胞P53的表达。结果 :通痹灵高低组、雷公藤高低组、青藤碱高低组、MTX高低组滑膜细胞凋亡的数量上明显高于造模组 ;P53蛋白的表达方面 ,造模组远远高于各组 ,与细胞凋亡的结果几乎相反。结论 :通痹灵、雷公藤多苷、青藤碱和MTX对RA滑膜细胞有较强的诱导凋亡作用 ,但并非通过对介导滑膜细胞P53的表达来诱导滑膜组织细胞凋亡。 Objective: To study the effects of Tongbiling, Tripterygium Glycosides and sinomenine on the apoptosis and P 53 expression of synoviocytes in collagen-induced arthritis (CIA) rats. Methods: Apoptosis level of synoviocytes in CIA rats of every group was tested by in situ cell death detection kit. The P 53 expression of synoviocytes was analyzed by immunohistochemistry technique. Results: As for the quantity of synoviocytes with apoptosis, the groups with high and low dosages of Tongbiling, Tripterygium Glycosides and sinomenine were obviously more than the model group; the P 53 expression of the models exceeded that of the other groups enormously, which is almost contrary to the result of apoptosis. Conclusion: Tongbiling, Tripterygium Glycosides and sinomenine have the effect to induce apoptosis of synoviocytes, but the apoptosis is not incuded by promoting P 53 expression of synoviocytes.
