摘要
目的研究莲威阿那其颗粒抗炎、软骨细胞增殖作用及安全性。方法将72只小鼠随机分为6组:模型组、阳性组(醋酸泼尼松0.09mg/g),水煎液组(处方药17.64mg/g),莲威阿那其颗粒低(2mg/g)、中(4mg/g)、高(8mg/g)剂量组。采用角叉菜胶致小鼠足跖肿胀实验,观察莲威阿那其颗粒的抗急性炎症作用。取大鼠膝关节软骨建立软骨细胞体外培养体系,采用MTT法进行检测,观察药物对大鼠软骨细胞增殖的影响。通过急性毒性实验,确定小鼠对莲威阿那其颗粒的最大耐受量(MTD)。结果与模型组比较,莲威阿那其颗粒水煎液组、阳性组可抑制角叉菜胶所致小鼠足趾肿胀度差异有统计学意义(P<0.01);莲威阿那其颗粒中、高剂量可抑制角叉菜胶所致小鼠肿胀度,差异有统计学意义(P<0.05)。与对照组比较,莲威阿纳其颗粒高剂量组可增殖大鼠软骨细胞,差异有统计学意义(P<0.01);中剂量组可增殖大鼠软骨细胞,差异有统计学意义(P<0.05);与模型组相比,低、高、中剂量组均可增殖大鼠软骨细胞,差异有统计学意义(P<0.01)。以莲威阿纳其颗粒每天50g/kg剂量灌胃给药7d小鼠全部存活,相当于成人临床用药量的116.67倍。结论莲威阿那其颗粒具有抗急性炎症、促进软骨细胞增殖的作用,观察期内未见明显毒性反应。
Objective To study the effect and safety of Lianweianaqi granules on inflammation and cartilage cell proliferation.Methods 72 mice were randomly divided into 6groups:model group,positive control group(prednisone 0.09 mg/g),decoction group(prescription 17.64 mg/g),Lianwei Anaqi granule low(2mg/g),middle(4mg/g),high dose group(8mg/g).Using mice metatarsal swelling model induced by carrageen glue to observe the anti-acute inflammation of Lianweianaqi granules.Taking the knee joint cartilage of rats to establish cells in vitro cartilage system,MTT method was used to test the effects of drugs on cartilage cell proliferation.The acute toxicity experiment was conducted to determine the largest drug tolerance(MTD).Results Compared with model control group,Lianweianaqi granules in water decoction group significantly inhibited the mice metatarsal swelling caused by carrageen glue with statistical signifi-cance(P <0.01),and Lianweianaqi granules in high dose and middle dose also inhibited the swelling with statistical significance(P <0.05).Compared with the model group,3doses groups were significantly increasing the proliferation of rat chondrocytes(P <0.01).All mice fed Lianweianaq particles with 50g/kg survived,and which is equivalent to 116.67 times of the adult clinical dose.Conclusion Lianweianaq particles had the obvious resistance to acute inflammation and promoted the chondrocytes proliferation,and there was no obvious toxic effects during observation period.
